Extensive experience in the identification of macromolecular interactions by site-directed photosensitized reactions. Monospecific antibodies and other protein ligands conjugated to visible light photosensitizers are powerful tools to demonstrate whether macromolecules and/or cellular organelles interact in situ and in vitro. Using this site-directed approach the expert has shown that DNA repair enzymes bind microtubules and explored the photodynamic therapeutic potential of antibody-porphyrin conjugates.
Extensive experience in developing immunoassays and in raising and screening polyclonal and monoclonal antibodies. Antibody production has included antibodies raised against proteins and low molecular mass molecules (less than 0. 5 kD). Extensive experience in customization of immunoassays, including enzyme linked immunosorbent assays (ELISA), immunoblotting (Western blots), immunoprecipitations, and affinity column chromatography. Using these approaches the expert has been able to detect in vitro and in situ the DNA lesion, 8-oxoguanine.
Extensive experience in the identification of molecular targets for naturally occuring toxins through combinations of mass spectral and proteomic analyses. Using LC/MS/MS the expert has been able to detect and quantitate DNA damage, as well as alterations in metabolic pathways, in small mammals exposed to toxins. In addition, she has developed assays by which immobilized toxin is used to pull down potential protein targets in tissue homogenates. Identification of protein targets is performed by combining biochemical, immunochemical and proteomic analyses. The expert also has detected and quantitated toxins from animal isolates and dietary supplements.
