Need molecular biology expert. For plasmid maxi prep, i started a 100 ml culture in n ampicillin lb broth from a 2 week old 2 ml lb-amp culture from 4 deg c. The 100 ml culture grew well overnight. I did not see a dna pellet after isopropanol step while i saw dna pellets from 3 other simultaneous maxi preps where 100 ml culture was inoculated from fresh 2ml cultures. All plasmids are high copy. I used qiagen endofree maxi prep kit. I need to know why i did not get dna pellet in first plasmid while the 100 ml culture grew well from old 2 ml culture.

We are setting qpcr to ampliphy few genes and using gabdh as internal control. We are using abi 7900 we get a very good expression value for gabdh, however for other il6 and il2, we get good rq and ddct for sample and other says is not detected although all the rna/cdna used and prepared at the same time. I have some clients who are interested in having their whole genome sequenced and finding out their genetic risk for various common disorders. They have already done the 23andme and decodeme snp analysis, and are in the process of doing the illumina trusight individual genome sequencing (igs) test with the mygenome ipad app frontend. Looking for a consultant with very broad knowledge of genomic sequencing field, knows the key companies and players involved, knows the software products availabe to analyze genomes, and can help me get this project started. I am interested in someone who knows the field so i don't have to reinvent the wheel, so i can pick the low hanging genomic analysis products available off-the-shelf before doing any expensive custom analysis of whole genomes for my clients.
